The denomination extends across the Valladolid and Segovia plateaux at altitudes of 700 to 900 metres, on soils with a topsoil of sands and gravel over clay and limestone. The continental climate, with cold winters and warm summers, and the cool nights of the plateau preserve the natural acidity of the grape during the ripening period.

Verdejo is the indigenous white variety and the axis of the denomination, with an aromatic profile of fresh herbs, fennel and almond, and a rich texture with structured bitterness on the finish. The DO requires a minimum of 50% Verdejo in wines carrying the denomination's name and 85% for those labelled Rueda Verdejo. Sauvignon Blanc and Viura act as complementary varieties; sparkling wine production is growing, although the bulk of production remains still, on lees.
